Potrich,Michele; Silva,Rita T.L. da; Maia,Fabiana M.C.; Lozano,Everton R.; Rossi,Robson M.; Colombo,Fernanda C.; Tedesco,Flavia G.; Gouvea,Alfredo de.
ABSTRACT This study aimed to evaluate the effect of commercially used entomopathogens on Africanized Apis mellifera L. (Hymenoptera: Apidae). Four bioassays were performed: 1) pulverized entomopathogens on A. mellifera; 2) entomopathogens sprayed on a smooth surface; 3) entomopathogens sprayed on soy leaves; and 4) entomopathogens mixed with candy paste (sugar syrup). Five treatments were prepared: sterile distilled water (control), distilled water sterilized with Tween® 80 (0.01%), and the commercial entomopathogens Metarhizium anisopliae E9 (1.0 × 109 conidia mL−1), Beauveria bassiana PL63 (1.0 × 108 conidia mL−1) and Bacillus thuringiensis var. kurstaki HD-1 (3.0 × 108 spores mL−1). Each treatment consisted of five repetitions, with 20 workers per...

Elasmopalpus lignosellus (Zeller, 1848) (Lepidoptera, Pyralidae) is an insect pest of 60 economically important crops, including sugarcane, wheat, soybean, rice, beans, sorghum, peanuts, and cotton. The aim of this work was to select and characterize Bacillus thuringiensis isolates with insecticidal activity against E. Lignosellus that could be used as an alternative method of control. Selective bioassays were done to evaluate the toxicity of 47 isolates against first instar larvae of E. lignosellus. For the most toxic bacterial strains, the lethal concentration (LC50) was estimated and morphological, biochemical and molecular methods were used to characterize the isolates. Among the 47 isolates tested, 12 caused mortality above 85% and showed LC50 values...

Isolation and molecular characterization of Bacillus thuringiensis found in soils of the Cerrado region of Brazil, and their toxicity to Aedes aegypti larvae Rev. Bras. entomol.
Lobo,Katiane dos Santos; Soares-da-Silva,Joelma; Silva,Maria Cleoneide da; Tadei,Wanderli Pedro; Polanczyk,Ricardo Antonio; Pinheiro,Valéria Cristina Soares.
ABSTRACT This study investigated the potential of Bacillus thuringiensis isolates obtained in the Cerrado region of the Brazilian state of Maranhão for the biological control of Aedes aegypti larvae. The isolates were obtained from soil samples and the identification of the B. thuringiensis colonies was based on morphological characteristics. Bioassays were run to assess the pathogenicity and toxicity of the different strains of the B. thuringiensis against third-instar larvae of A. aegypti. Protein profiles were obtained by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Polymerase chain reaction assays were used to detect the toxin genes found in the bacterial isolates. Overall, 12 (4.0%) of the 300 isolates obtained from 45 soil samples were...

Isolation, morphological and molecular characterization of Bacillus thuringiensis strains against Hypothenemus hampei Ferrari (Coleoptera: Curculionidae: Scolytinae) Rev. Bras. entomol.
Zorzetti,Janaina; Ricietto,Ana Paula Scaramal; Fazion,Fernanda Aparecida Pires; Meneghin,Ana Maria; Neves,Pedro Manuel Oliveira Janeiro; Vilas-Boas,Laurival Antonio; Vilas-Bôas,Gislayne Trindade.
ABSTRACT The coffee berry borer Hypothenemus hampei Ferrari, 1876 (Coleoptera: Curculionidae: Scolytinae) is considered the most serious pest of the coffee crop and is controlled primarily with the use of chemical insecticides. An alternative to this control method is the use of the entomopathogenic bacterium, Bacillus thuringiensis Berliner, 1911. Therefore, the objective of this work was to select strains of B. thuringiensis virulent against H. hampei and characterize them by morphological and molecular methods to identify possible genes for the production of genetically modified plants. To achieve this objective, 34 strains of B. thuringiensis underwent a selective bioassay to evaluate their toxicity to H. hampei first-instar larvae. Among the strains...

Molecular characterization of Bacillus thuringiensis strains to control Spodoptera eridania (Cramer) (Lepidoptera: Noctuidae) population Rev. Bras. entomol.
Machado,Déborah Heloísa Bittencourt; Livramento,Kalynka Gabriela do; Máximo,Wesley Pires Flausino; Negri,Bárbara França; Paiva,Luciano Vilela; Valicente,Fernando Hercos.
Abstract The main objective of this study was to characterize the toxicity and genetic divergence of 18 Bacillus thuringiensis strains in the biological control of Spodoptera eridania. Bacterial suspensions were added to the S. eridania diet. Half of the selected B. thuringiensis strains caused high mortality seven days after infection. The genetic divergence of B. thuringiensis strains was assessed based on Enterobacterial Repetitive Intergenic Consensus (ERIC) and Repetitive Extragenic Palindromic (REP) sequences, and five phylogenetic groups were formed. Despite their genetic diversity B. thuringiensis strains did not show any correlation between the collection sites and toxicity to larvae. Some B. thuringiensis strains are highly toxic to S. eridania,...

Mortality of Oryzophagus oryzae (Costa Lima, 1936) (Coleoptera: Curculionidae) and Spodoptera frugiperda (J E Smith, 1797) (Lepidoptera: Noctuidae) Larvae Exposed to Bacillus thuringiensis and Extracts of Melia azedarach BABT
Berlitz,Diouneia Lisiane; Azambuja,Aline Oliboni de; Sebben,Alessandra; Oliveira,Jaime Vargas de; Fiuza,Lidia Mariana.
Oryzophagus oryzae (Costa Lima 1936) (Coleoptera: Curculionidae) and Spodoptera frugiperda (J E Smith, 1797) (Lepidoptera: Noctuidae) cause important crop losses in southern Brazil. Control is possible by the use of the bacteria Bacillus thuringiensis and extracts of Melia azedarach. This study aimed to evaluate the mortality, in vivo, of O. oryzae and S. frugiperda submitted to two isolates of B. thuringiensis and the aqueous extract of M. azedarach. The LC50 for O. oryzae due to bacteria was 5.40μg/mL (Bt 2014-2) and due to plant extract 0.90μg/mL. For S. frugiperda, the Bt 1958-2 bacterial suspension (1.10(10)UFC/mL) caused a 100% of corrected mortality, showing that the purified Cry proteins caused a CL10 of 268μg/mL five days after the treatments, and...
